PTP for T2DM mice serum metabolomics

Serum samples from four experimental groups (CON, T2DM, POS, and PTP-H; n = 7 per group) were subjected to untargeted metabolomics analysis by LC-MS/MS (Beijing Novogene Co., Ltd., Beijing, China) to investigate the systemic metabolic changes induced by PTP intervention in T2DM mice. Serum metabolites were extracted with 80% methanol aqueous solution, and quality control (QC) samples were prepared by mixing equal volumes of all serum samples to monitor instrument stability. Metabolites were separated on a Hypesil Gold C18 column and detected on a Q Exactive HF mass spectrometer in both positive and negative ion modes. Raw data were processed using Compound Discoverer 3.1, and metabolites were identified against the mzCloud, mzVault, and MassList databases. Differential metabolites were screened using VIP > 1, P < 0.05, and FC ≥ 2 or FC ≤ 0.5, and functional annotation was performed using the KEGG, HMDB, and LIPIDMaps databases. This dataset provides a comprehensive profile of serum metabolic alterations associated with PTP-mediated improvement of glucolipid metabolism in T2DM mice.
