Description

The data displayed 390 Differential metabolites (DMs) up-regulated (Col-0-LP vs fer-4-LP, VIP>1, FC>1.5, P<0.05) in the positive ion mode and 483 DMs up-regulated in the negative ion mode.

Steps to reproduce

To sufficiently collect the metabolites in roots in vitro, fifty seedlings of Col-0 or fer-4 growing on LP medium for 10 days were harvested into 2 mL tubes. After grinding in liquid nitrogen, seedlings were added with aqueous methanol acetonitrile solution (2:2:1, v/v), vortexed and sonicated twice, then left at -20°C for 1h to precipitate the protein. After filtering, the supernatant was centrifuged at 14000 g for 20min at 4°C, and the new supernatant was freeze-dried and the samples were stored at -80°C. Aliquots of samples were mixed as QC samples. Metabolite analysis was carried out using the Agilent 1290 Infinity LC UHPLC system (Agilent Technologies, USA) in combination with the AB Triple TOF 6600 Mass Spectrometer (AB SCIEX, USA). Metabolite structures were identified using the method of accurate mass number matching (<25 ppm) and secondary spectrum matching, and then, were searched from the self-built database of Shanghai aptbiotech Co., Ltd. (Shanghai, China).
