Description

This dataset contains the raw and processed data supporting the findings of the study titled "MaARF18-like-2 Suppresses Low-Potassium Tolerance in Banana by Repressing MaWRKY22-X1 Transcription and Disrupting Cellular ROS Homeostasis" by Wei et al. The data are derived from experiments investigating the regulatory role of the banana ARF transcription factor MaARF18-like-2 in low-potassium stress tolerance, including its transcriptional repression of MaWRKY22-X1 and its impact on cellular reactive oxygen species (ROS) homeostasis. File List and Descriptions: Figure_1_original_data.zip: Contains all data related to the phylogenetic tree construction. Figure_2_original_data.xlsx: Contains processed expression data used for generating the tissue-specific expression patterns of MaWRKYs. The data include:Relative expression levels of MaWRKYs across different banana tissues (e.g., root, stem, leaf, flower, fruit).Normalized expression values calculated from Ct values using the 2^−ΔΔCt method. Experimental Methods Summary Genome-wide identification and phylogenetic analysis: WRKY family members were identified from the banana genome database. Phylogenetic trees were constructed using MEGA with maximum likelihood method (bootstrap = 1000 replicates). qPCR expression analysis: Total RNA was extracted from banana tissues and subjected to reverse transcription. qPCR was performed using gene-specific primers, with MaActin as the internal reference gene. Relative expression was calculated using the 2^−ΔΔCt method. Dual-luciferase reporter assay: The promoter of MaWRKY22-X1 was cloned into the LUC reporter vector, and MaARF18-like-2 was cloned into the effector vector. LUC/REN ratios were measured using a dual-luciferase assay system to evaluate transcriptional repression activity. Arabidopsis transgenic lines: MaARF18-like-2 and MaWRKY22-X1 was overexpressed in Arabidopsis via Agrobacterium-mediated transformation. Transgenic lines were selected and subjected to low-potassium stress treatment for phenotypic and physiological characterization.
