Transcriptomic Analysis of Chinese Longevity Cohorts

Description

A total of 1,751 individuals, comprising 811 long-lived individuals (LLIs) and 940 younger controls (YCs), were recruited. Peripheral blood was drawn, and white blood cells were isolated for total RNA extraction using the TRIzol method. The RNA-seq libraries were prepared using either polyadenylate capture or rRNA depletion methods, followed by sequencing on Illumina high-throughput platforms. We analyzed transcriptomes from these LLIs and YCs to map transcriptional aging trajectories. Raw reads were mapped to hg19 human reference genome. The aligned reads were annotated using the matched RefGene database. Gene-level read counts for protein-coding genes were generated with the tool featureCounts.
