Metadata for "Effects of Short-Term Warming and Grazing on Soil Microbial Communities in Saline-Alkaline Grasslands of Northern China"
Description
This dataset contains the raw sequencing data and complete metadata for the manuscript entitled "Effects of Short-Term Warming and Grazing on Soil Microbial Communities in Saline-Alkaline Grasslands of Northern China" (Manuscript ID: 1834208). Raw sequencing data: - 16S rRNA gene amplicon sequencing data (V3–V4 region, 32 samples, paired-end fastq files) - ITS region amplicon sequencing data (ITS1 region, 32 samples, paired-end fastq files) The metadata file includes: - Sample ID and treatment grouping (grazing intensity: NG, no grazing; LG, light grazing; MG, moderate grazing; HG, heavy grazing; warming treatment: CK, ambient control; W, +2°C canopy warming) - Block information (Block 1–4) - Soil physicochemical properties: pH, electrical conductivity (EC, µS/cm), dissolved organic carbon (DOC, mg/kg), dissolved nitrogen (DN, mg/kg), DOC/DN ratio - Soil water content (SWC, %, volumetric) measured in situ using a portable TDR soil moisture meter (TDR-350, Spectrum Technologies, USA) twice monthly from May to August 2024 - NMDS coordinates for bacterial and fungal communities (based on Bray-Curtis distances) - Alpha diversity indices (Chao1, Shannon, Simpson, Pielou) for bacterial and fungal communities at the ASV level Study site: Youyu Loess Plateau Grassland Ecosystem Research Station (112°19′E, 39°59′N, elevation 1348 m), Shanxi Province, China. Soil type: Chestnut calcareous soil (Chinese soil classification), soil pH 9–10, with obvious saline-alkaline characteristics. All soil samples were collected in August 2024 from the 0–10 cm soil layer. Each sample is a composite of six soil cores (5 cm diameter) collected from the central core area (approximately 4–5 m²) of each subplot to minimize edge effects. Grazing treatments have been applied annually since 2017 (June 1 to September 30), and warming treatments since May 2023. Sequencing was performed on the Illumina NextSeq 2000 platform (Illumina, San Diego, CA, USA) by Shanghai Majorbio Bio-Pharm Technology Co., Ltd. (Shanghai, China).
