Base pair resolution chromosome conformation capture reveals mechanistic insights into the nature of chromatin structure

Description

This repository contains Micro Capture-C ultra data from the gene promoters of Cbfa2t2, Hba-a1 & Hba-a2, Hbb-b1, Idh2, Klf4, Med13l, Myc, Nanog, Setdb1, Slc25a37 (mitoferrin), Sox2, Tet2 and Trp53. 5 enhancers are included at these loci (Hba-a1, Myc, Nanog, Sox2 and Tet2) as well as 20 additional enhancers bound by SOX2. The following experimental conditions are included: wildtype E14 mouse ES cells (E14-WT), Ter 119+ erythroid cells (Ery-WT) and haematopoietic stem and progenitor cells (HSC-WT). Data are included from experiments in mouse ES cells with FKBP12(F36V) dTAG degrons for Mediator complex components MED1 (M1dTAG), MED13 (M13dTAG) and MED14 (M14dTAG). In addition, data are included for FKBP12(F36) dTAG degrons in mouse ES cells for SOX2 (SOX2dTAG) and NANOG (NanogdTAG). All degron experiments were conducted 2 hours post addition of dTAG13. These heatmaps represent data combined from all replicates. At least 6 replicates in total are included for each heatmap (3 biological (each with 2 technical replicates)). The heatmaps labelled represent the strength of contacts. The coordinates of all heat maps are included in the excel spreadsheet. A word document is included with additional figures that were generated in response to reviewer comments.
