Sampling was conducted in the main channel of the Weihe River and in the Fenghe, Bahe, and Chanhe Rivers. Each river was divided into three distinct sections: upstream, midstream, and downstream, as shown in the schematic diagram (Fig. 2b). Three consecutive rapid-pool-benchland systems were identified within each river section. One representative sampling site was established in each habitat within each system (Fig. 2c and d). This design resulted in a total of 108 samples for the entire study (4 rivers × 3 sections × 3 systems × 3 samples per system). During sampling, the surface sediment was removed. Using a grab sampler, multiple grabs were taken around the central point of the representative sampling site in the rapid, pool and benchland areas. Fresh sediment from 1 cm to 10 cm depth was collected using a core sampler and mixed into a composite sample. Simultaneously, water samples were collected directly above the sediment collection sites and stored in 500 mL polyethylene bottles. All samples were preserved at 0–4°C. A small amount of sediment from each sampling site was stored in PowerBead Pro Tube ‌at -40°C for DNA extraction, while the remainder was used for physicochemical analysis.
