Steps to reproduce
Lentiviral transduction of Brie library into Cas9-expressing CAD5 cells (undifferentiated state) at MOI of 0.3 followed by selection with puromycin for >1 week. Genomic DNA extraction of library cells at 500x representation with Qiagen Blood and Cell Culture DNA Midi Kit followed by PCR amplification sgRNA regions and appendage of Illumina barcodes and stagger regions. Illumina NextSeq 500 high-output 75pb run with 10% Phi-X spike-in. This is raw .fastq file from run.
