Steps to reproduce

BioID-based proximity labeling The BioID proximity labeling assay was conducted. Briefly, 293T cells were transfected with plasmid synthesized with bait gene (GPR35) and fused with BirA*. Then the proteins nearby GPR35 were biotinylated by BirA* enzyme and isolated using streptavidin affinity purification and identified via liquid chromatography-mass spectrometry (LC-MS/MS) analysis. . A fusion protein of human GPR35 with a HA-tag (YPYDVPDYA-tag), and a mutant form of biotin ligase (BirA*) were constructed by Creative-Biolabs and transfected into HEK293T cells. The 508 proximity interactors are shown with SAINT score (confidence score of protein-protein interaction, scale 0-1 with 1 being strongest) greater than 0.8.
