Malaria monoclonal antibodies in the humanized liver mouse model: dose to serum concentration relationship and protection afforded by pre-erythrocytic and erythrocytic stage mAbs

This work aims to advance preventative strategies for malaria. The study evaluated the protection afforded by passive transfer of a pre-erythrocytic antibody (mAb 317), a blood stage monoclonal antibody (mAb RH5), or both. This dataset consists of results for 278 humanized liver mice (FRGhuHep mice) infected with Plasmodium falciparum by mosquito bite, with administration of monoclonal antibodies on Day (-1) and/or Day 5 as indicated. Administered dose in ug and serum concentration in ug are provided for days indicated. Parasitemia results in parasites/mL as measured by qRT-PCR at D7-13 following infection are provided. Not all fields are relevant for each experimental subject (e.g., pre-erythrocytic-only experimental conditions do not have Day 5 mAb dose, D6 serum concentration, or D11/D13 parasitemia results). The overall result is indicated, defined as "positive" if any collected blood sample measured a parasitemia level of 5 parasites/mL or greater on any day of blood collection.

Steps to reproduce
FRGhuHep mice were administered antibody at indicated days, infected with P. falciparum by mosquito bite (D0), administered human red blood cells (D5, D6, +/- D9), and samples collected (D7, D9, +/- D11 and D13). Serum samples were collected on indicated days. Additional RBC injections and collections are for some experiments conducted testing erythrocytic stage mAb (RH5). Blood samples were analyzed by qRT-PCR for P. falciparum 18S RNA. Serum samples were evaluated for presence of human IgG. Variables in dataset: mAb: identity of the monoclonal antibody administered Dose in ug: mAb administered at D(-1) Dose in ug: mAb administered at D5 D0 serum concentration in ug D6 serum concentration in ug D13 serum concentration in ug Sample ID (D7): this is an internal identified of the sample. D7 is indicated, as D9, D11, and D13 samples have unique sample IDs but are provided in this dataset on the same line, as one line represents one mouse. D7 parasitemia (par/mL) D9 parasitemia (par/mL) D11 parasitemia (par/mL) D13 parasitemia (par/mL) Overall Result: POS/NEG indicates the overall result, where POS indicates any sample at 5 parasites/mL or greater, and NEG indicates all samples below 5 parasites/mL.
