Steps to reproduce
H9c2 cells were growth in Advanced Dulbecco's modified Eagle's medium (Thermo Fisher Scientific, Waltham, MA, USA), supplemented with 2 mM L-glutamine and 5% fetal bovine serum-FBS. After confluence cells were activating by grow in high glucose (30.0 mM) medium. Cells were also activated by Digoxin and Ouabain, both with 1.0 mmol/L for 30 minutes. Extraction was performed using urea/thiourea buffer, followed by tryptic digestion and analyzed in an Orbitrap mass spectrometer by LC-MS/MS. Raw data files were first analyzed by Sequest and them the .msf files were loaded in the Scaffold software and quantified by Total spectra method using T-Test with p<0.05 (y axis: -log10 p-value) and at least three fold change (x axis: log2 of fold change).
