Overexpressed Spindlin1 relocalizes to the nucleoli during fixation

Description

To quantify the relocalization of overexpressed Spindlin1 during fixation, confocal images have been evaluated according to fluorescence intensity within the nucleoli in comparison to the surrounding nucleuplasm: Relative fluorescence intensity of nucleoli and nucleoplasm was measured using ImageJ 1.54f. The nucleoplasm without nucleoli and the nucleoli were manually outlined as regions of interest (ROI). Where hard to delineate, the corresponding phase contrast image or Hoechst-channel was used as guide. The pixel values were averaged for each ROI. Where several nucleoli per cell were measured, the respective ROI mean values were averaged. This data set includes evaluated raw data from experiments analysing fixation temperature, formaldehyde concentration and live fixation.
