Description

This study is the first to report the identification of L. plantarum at the strain level. Through WGS-SNP analysis, three sets of primer–probe sequences were established (Table S1). The specificity and sensitivity tests demonstrated that all three sets of primers and probes successfully enabled the identification of strain YZH81 (Table S2). To evaluate the applicability of this WGS-SNP primer design methodology, we subsequently designed two additional sets of primer–probe sequences (Table S1) for other laboratory-preserved L. plantarum strains, WCFS1 and ST-III. The specificity and sensitivity experiments (Figure S1) confirmed successful detection of the respective strains. In summary, we successfully established a primer–probe design method based on WGS-SNP analysis for the strain-level identification of L. plantarum. Subsequently, the first set of primers and probes for YZH81 (Table 3) was selected for further experiments.
