In this research, we demonstrate how gene architecture directs the splicing outcome in terms of exon and intron definition in the nuclear spatial regions.

Steps to reproduce
Constructed minigenes from the leveled and differential architectures were stably expressed in 293HEK cells Flp-In System in genomic sites one that is found in the center of the nucleus and the other in the periphery. RNA extract was performed and cDNA using Reverse Transcription PCR. PCR was performed using primers specific to the UTR sequence of the plasmid (upstream and downstream of the minigenes). PCR reactions were run on 1.5% agarose gel electrophoresis. Ladder: 100bp DNA Ladder H3 RTU (GeneDireX, Inc.)
