Description

Original western blot images of fibronectin (Abcam, ab2413) and alpha-actinin (Sigma-Aldrich, A7811) staining of healthy, dystrophic and severely dystrophic muscle lysates (males, 11 weeks, n=5/mouse model). First lane (left lane) include Cell Signaling Technology prestained protein marker (10-250 KDa, 74124S). Last lane (right lane) include Thermo multicolor protein ladder (40-300 KDa, 11872124) . For simplicity only left lane standard is shown in the image.

Steps to reproduce

Western blot was performed according to manufacturer instructions (Cell Signaling Technology, 12957S). 4-20% protein gels (Bio-rad, 3450033) were used to load samples. Transfer was carried out in wet conditions for 120 minutes at 200 mA in cold room. Images were obtained using a ChemiDoc MP Imaging system. ImageLab (v5.1 and 6.1) was used for densitometric analysis of FN and alpha-actinin. Fold change to alpha-actinin and one-way ANOVA with Tukey's multiple comparisons were carried out using Graphpad Prim (v 9.1.1).
