Determining the Radiobiological Effects of Clinically Relevant Radiation Dose and Modality in Cervical Cancer

Description

The data in these files was collected to study the effects of radiation on cervical cancer cell lines. We examined the clonogenic survival, DNA repair capacity via gamma-H2AX foci induction and repair, and cytokine secretion following the irradiation by an orthovoltage pre-clinical irradiator, high dose rate brachytherapy remote afterloader, and clinical c-arm style linear accelerator. The alpha/beta ratios were generated using only the pre-clinical irradiator. All three irradiators were utilized (separately) for the single fraction multi-modality irradiation, gamma-H2AX, and cytokine profiling data. Colonies from the clonogenic data was manually counted using a microscope. Gamma-H2AX foci were counted using CellProfiler where each condition was given its own optimization parameters. This automated counter was validated against manual counting of the same images. The cytokine data was generated from a multiplex immunosorbent assay ran on a fluorescent flow cytometry based Luminex instrument. The conditioned media for this was collected after a 24hr incubation. Each irradiation and modality was given its own non irradiated sham group. The experimental setup essentially requires this as each irradiator is located in a separate space and thus have different conditions (walk time, etc.).
