Steps to reproduce

Figure 1: Gly, D-Ser, or L-Glu was applied to HEK293 cells expressing various combinations of GluD1, GluD2, Cbln1, and Nrxn, or none of these, while being held at -60 mV. Figure 2: Gly was applied via a theta tube to HEK293 tSA cells expressing GluD2, GluD2-dATD, or GluA2, while being held at -60 mV. Figure S1: Gly or Gly+NASPM was applied to HEK293 tSA cells expressing none or GluD2/Cbln1/Nrxn1, while being held at -60 mV. Figure S2: HEK293 tSA cells were immunostained with an anti-HA antibody under non-permeabilized conditions. Confocal images were acquired using an Olympus FLUOVIEW FV1000 microscope. Figures 3 and S3: Gly or L-Glu was applied with or without NASPM, NBQX, or sarcosine to whole-cell voltage-clamped Purkinje cells (-70 mV) in cerebellar slices acutely prepared from wild-type or GluD2-null mice.
