Description

This dataset contains the source and derived data underlying Figures 1–6 and Supplementary Figure S2 of the related manuscript. The electrophysiological data include per-slice field excitatory postsynaptic potential (fEPSP) amplitude and slope time courses recorded at Schaffer collateral–CA1 synapses in hippocampal slices from adult male rats, together with the values used to quantify long-term depression during the final 10 min of post-LFS recording. The behavioral data include daily exploration measurements from the prolonged novel object recognition paradigm and per-animal discrimination indices for Days 2, 4, and 6. Data are provided as machine-readable CSV files, accompanied by the original GraphPad Prism projects containing the statistical analyses and a README describing the files, variables, units, experimental timing, group codes, discrimination-index calculations, and quality-control procedures.

Steps to reproduce

Download and extract the dataset archive and consult README.txt for file definitions, experimental timing, group codes, and data-processing details. To reproduce the electrophysiological time-course plots, import fig1-5_fEPSP_timecourse.csv, group observations by treatment and sample_id, and plot amplitude_percent_of_baseline or slope_percent_of_baseline against experimental_time_min. Blank fEPSP fields during experimental time 30–70 min correspond to the omitted LFS period. Use fig1-5_quantification_140-150min.csv for the final-window LTD analyses. Analyze amplitude and slope separately. For Figure 1, use ordinary one-way ANOVA followed by Dunnett’s multiple comparisons test versus control. For Figures 2 and 5, use ordinary one-way ANOVA followed by Tukey’s multiple comparisons test. For Figures 3 and 4, use an unpaired Student’s t-test. To reproduce the daily NOR plots and within-session comparisons in Figure 6B–H and Supplementary Figure S2A, use percentage_of_total_prism in fig6_S2_exploration_times.csv. Match the two object observations by animal_id, group, and day, and perform a paired Student’s t-test separately for each group and session without correction across sessions. To reproduce the discrimination-index analyses, use the exact discrimination_index values in fig6_S2_discrimination_index.csv. Analyze Day 2 and Day 4 separately by ordinary one-way ANOVA followed by Dunnett’s multiple comparisons test with saline as the control. Analyze Day 6 by ordinary one-way ANOVA followed by Tukey’s multiple comparisons test. The recalculated DI fields are provided only for auditing and should not replace the exact Prism values. The original analysis settings, statistical outputs, and graphs are retained in prism/LTD.prism and prism/NOR.prism. The projects were saved using GraphPad Prism for macOS.
