Description

Soluble Biomarkers detected by Olink in the serum of children with multisystem inflammatory syndrom (MIS-C), Kawasaky disease (KD) or Still's disease (SD) before treatement as well as healthy children (HC) and convalescent COVID-19 (CC). The MIS-C group included 22 pediatric patients sampled at diagnosis, before intravenous immunoglobulin or corticosteroid treatment, at a median of 5.5 weeks [IQR 3.1–7.8] after SARS-CoV-2 infection. MIS-C was diagnosed according to WHO, CDC, and Royal College of Paediatrics and Child Health criteria 22–24. The CC group included 17 children sampled during COVID-19 convalescence, at a median of 3.7 weeks [IQR 3.1–4.7] post-infection and served as the MIS-C control group. Both cohorts have been previously described25 . The KD group included 15 pre-treatment pediatric patients recruited before the SARS-CoV-2 pandemic, avoiding overlap with MIS-C, and diagnosed according to American Heart Association criteria26 . The SD group included 8 patients at first disease onset, recruited before and after the SARS-CoV-2 pandemic, sampled before therapy, and fulfilling ILAR criteria for systemic juvenile idiopathic arthritis18. The HC group included 15 healthy children aged 2–13 years, recruited before the pandemic, and served as controls for KD, and SD.

Steps to reproduce

Serum samples from multisystem inflammatory syndrome in children (M), Kawasaki disease (K), children onset Still's disease (JIA), pediatric convalescent covid (C), and healthy children (HC), were analyzed using the Olink® Target 96 Inflammation panel, which quantifies 92 inflammation-associated proteins using proximity extension assay technology. Samples were processed on a single assay plate in single technical replicates. Data were normalized using internal controls, and triplicate negative controls were used to define background signal and limits of detection. Protein levels are reported as normalized protein expression values on a log₂ scale.
