Dose Response Test of Patient - derived Cancer Organoids to Irradiation

Our research hypothesis was that patient-derived rectal cancer organoids would exhibit dose-dependent radiosensitivity to X-ray irradiation (0–16 Gy) and that image-based analysis would more accurately reflect this sensitivity than the CCK8 assay. The dataset includes two key files: radiation survival.pzf (image-based survival counts of organoids) and radiation cck8.pzf (CCK8 assay absorbance values measured via microplate reader, with 3 biological replicates per dose/time point). The data shows organoids displayed tumor-like heterogeneity and a clear dose-dependent decrease in survival (e.g., 16 Gy reduced survival to ~15% of controls via imaging), while the CCK8 assay failed to accurately capture this response; a notable finding is that image-based analysis generated representative survival curves (with a linear slope, shoulder region, and subsequent straightening), confirming its reliability for assessing organoid radiosensitivity. This data can be interpreted as validating rectal cancer organoids as a viable model for radiosensitivity research and image-based analysis as the optimal assessment method. To use it, researchers can apply tools like Excel or GraphPad Prism for replicating survival curve analyses or secondary research (e.g., exploring sub-group differences in organoid sensitivity), with all experiments repeated and outliers excluded via Grubbs’ test to ensure reproducibility.

Steps to reproduce
The dataset was generated through the following workflow: Fresh surgical specimens of primary rectal carcinoma were collected from patients treated at the Department of Gastrointestinal Surgery, Union Hospital, Tongji Medical College, Huazhong University of Science and Technology—all patients received no treatment before radical rectal cancer resection, with informed consent obtained from each participant, and all specimen collections and experiments approved by the Institutional Review Boards of the same hospital. Patient-derived rectal cancer organoids were established from these specimens, with genetic characterization via targeted sequencing to confirm tumor heterogeneity. Organoids were then exposed to gradient X-ray doses (0–16 Gy) using a linear accelerator, followed by survival assessment via two methods: image-based analysis (captured with an inverted microscope and analyzed for intact organoid counts) and CCK8 assay (absorbance measured at 450 nm using a microplate reader, with 3 biological replicates per dose/time point). Raw data were processed in Excel and GraphPad Prism, with outliers excluded via Grubbs’ test and experiments repeated to ensure reproducibility; key reagents included trypsin-EDTA, collagenase IV, Matrigel, organoid culture supplements, and CCK8 kits.
