This study investigated the time-dependent circulating levels of multiple chemotherapeutic agents using an in ovo chick embryo CAM model coupled with targeted UPLC-PRM-MS/MS analysis. Following controlled embryonic development, drugs were administered through the CAM vasculature, and blood samples were collected at predefined time points. Samples underwent analyte-specific extraction and, where required, chemical derivatization before LC–MS/MS analysis. Drug signals were evaluated against spiked reference and untreated control samples to obtain relative circulating levels and establish background limits, followed by statistical comparison of temporal changes.
