Description

WT vs KO: Untargeted lipidomic profiling on livers of Wildtype and Adipocyte specific Isthmin1 knockout mice to determine the specific lipid species regulated by adipose-derived Ism1 after 6 weeks on the MASLD diet. Saline vs FXR and Saline vs ISM: WT mice with established MASLD were dosed I.P. daily with either vehicle, 5 mg/kg Ism1, or GW4064 FXR agonist at 30 mg/kg for 2 weeks.

Steps to reproduce

Lipid samples were transferred into MS vials and 10-20 µL were injected for MS analysis. Mass spectrometry analysis of lipids was performed with an Agilent 6470 Triple Quadrupole (QQQ). For separation of metabolites, normal-phase chromatography was performed with a Luna 5 µm C5 100 Å LC column. The buffers were as follows: A2, 95 water/5 methanol; B2, 60 isopropanol/35 methanol/5 water. The additives were as follows: 0.1% formic acid each buffer (positive mode) and 0.1% ammonium hydroxide each buffer (negative mode). The flow rate started at 5 % B for 0-5 min, followed by 95 % B, flushed at 95 % B for 5-10 min, and requilibrated to 5 % B for 2-5 min. The acquired lipid species were determined using metlin.scripps.edu.
