Mitigating Colibactin Production and Genotoxicity

We hypothesized that colibactin production and its genotoxic effects could be mitigated to decrease its genotoxicity against human cells. We demonstrate that a safe dietary phytochemical has inhibitory activity against colibactin production using complementary approaches including a ClbP peptidase activity probe, clb transcriptional reporter assay and qPCR, DNA interstrand crosslinking assays, γ-H2AX induction (early DNA damage marker), megalocytosis, and β-gal senescence assays in human cell lines (Caco-2 and HeLa), providing robust evidence for hallmarks of genotoxicity. Data from these experiments are included in this data set.
