Steps to reproduce

The rice genotypes Varadhan (efficient) and Prasanna (non-efficient) used in the present study were selected based on their contrasting NUE response. The seeds of Varadhan and Prasanna were surface sterilized with 1% sodium hypochlorite and germinated in Petri plates lined with wet filter paper under dark conditions. After two days, the germinated seedlings were transferred to the plant growth chamber and maintained for 10 days (16 hours light/ 8 hours dark, 24°C). The 12-day-old uniform seedlings were transferred to trays containing Yoshida nutrient solution following the randomized complete block design with two replications. The two hydroponics treatments viz., Yoshida nutrient solution with recommended N source (N+) and without N source (NH4NO3) (N-) were maintained at standard growth conditions in the plant growth chamber. The hydroponics nutrient solution was renewed every seven days. The shoot and root samples were harvested on the 34th day for morphological and physiological observations and also for RNA isolation for gene expression analysis through microarray.
