Description

This dataset supports a study on engineering optimized matriptase substrates for protease-activated prodrugs. "Engineering high efficiency matriptase substrates using E. coli display for applications in prodrug activation". Anna Mestre Borras, Hanna Mehari, Stefan Ståhl and John Löfblom, doi: 10.1016/j.crmeth.2025.101077. A biased peptide library was displayed on E. coli and screened over four rounds using FACS after matriptase treatment. Files include raw and annotated sequences (original + Sort1–Sort4) from NGS, as well as clustered variants. The data reveal enrichment of efficiently cleaved sequences and were used to guide downstream kinetic and prodrug activation assays. Sequencing data were processed to extract substrate motifs, identify high-frequency hits, and assess cleavage preferences.
