润色后中文: 菌株NI-4分离自甘肃省武威市的羌活(Notopterygium incisum)种子内部。前期实验结果表明,该菌株是一株多功能植物促生菌,具备耐盐碱、产生吲哚-3-乙酸(IAA)等促生特性。全基因组分析显示,NI-4基因组由一条染色体和两个质粒拼接得到,三者均为环状结构。基于全基因组序列,预测到多个与植物生长促进相关的功能基因簇,涉及溶磷、固氮、IAA合成以及耐盐碱机制(如离子转运、氧化应激响应和相容性溶质合成)。 英文翻译: The bacterial strain NI-4 was isolated from the interior of Notopterygium incisum seeds collected in Wuwei City, Gansu Province. Previous experimental results demonstrated that NI-4 is a multifunctional plant growth-promoting bacterium, exhibiting traits such as salt-alkali tolerance and indole-3-acetic acid (IAA) production. Whole-genome analysis revealed that its genome consists of one chromosome and two plasmids, all circular in structure. From the genomic data, multiple gene clusters associated with plant growth promotion were predicted, including those involved in phosphate solubilization, nitrogen fixation, IAA biosynthesis, and salt-alkali tolerance mechanisms (e.g., ion transport, oxidative stress response, and compatible solute synthesis).

Steps to reproduce

Bacterial genomic DNA was extracted using the NucleoBond® HMW DNA kit (MN NucleoBond, Germany, Cat. 740160.20). The 16S rRNA gene was amplified with universal primers 27F (5'‑AGAGTTTGATCCTGGCTCAG‑3') and 1492R (5'‑GGTTACCTTGTTACGACTT‑3'). The PCR product was examined by pulsed‑field gel electrophoresis on a 0.7% agarose gel to verify its fragment size and purity. The final product was sequenced by Sangon Biotech (Shanghai) Co., Ltd. Raw sequencing data for the NI‑4 genome were obtained by combining de novo sequencing with both second‑generation and third‑generation sequencing platforms. Following quality control of the raw data, clean reads were de novo assembled and iteratively polished to generate a complete, high‑accuracy, and high‑integrity genome sequence of NI‑4.
