Per-animal raw data, Western-blot source data, and reagent inventories supporting "Phytochemically Standardised Aqueous Hellenia speciosa Rhizome Extract is Associated with Attenuated HFD/STZ-Induced Diabetic Hepatic Injury in Male Wistar Rats"
Description
This dataset provides the per-animal raw values, Western-blot source-data files, histopathology source images, HPLC-DAD raw chromatograms, reagent inventories, and statistical-analysis outputs underlying the summary statistics reported in the manuscript referenced below. The study evaluated a phytochemically standardised aqueous rhizome extract of Hellenia speciosa (J.Koenig) S.R.Dutta (synonyms: Costus speciosus; Cheilocostus speciosus; Costaceae; abbreviated CS-AE) in a six-arm preclinical design (n = 10 male Wistar rats per group): NC, NC + CS-AE, DC, DC + Metformin, DC + CS-AE, DC + MET + CS-AE. Diabetes was induced by 4 weeks of high-fat diet followed by a single low-dose streptozotocin injection (35 mg/kg). The six-week treatment phase quantified metabolic biochemistry, hepatic oxidative stress, ex vivo substrate-supported mitochondrial bioenergetics, Western-blot densitometry of the Nrf2/Keap1, AMPK/PGC-1a, mitochondrial-dynamics, and apoptosis axes, qRT-PCR (single-reference Gapdh plus dual-reference Gapdh + Hprt1 sensitivity re-analysis), NAS-adapted histopathology, Masson's trichrome morphometry, and immunohistochemistry for nuclear Nrf2, HO-1, and cleaved caspase-3. The deposit is structured as follows: 01_Per_Animal_Data: Supplementary Dataset S1 (15 worksheets) 02_Western_Blot_SourceData: full-membrane TIFFs, per-target densitometry, ROI overlays, processing methods 03_Histopathology_Photomicrographs: 5 stains x 60 animals x 5 fields per animal (1500 source images) 04_HPLC_Chromatograms: Agilent ChemStation .D folders for each batch, stability time-point, and reference standard 05_Inventories: antibody and TaqMan inventory spreadsheets 06_Statistical_Outputs: GraphPad Prism .pzfx, R scripts, ANOVA / ART-ANOVA / Spearman / geNorm output PDFs 07_qPCR_Raw_Ct_Values: QuantStudio .eds and .xlsx exports ARRIVE 2.0, ICH Q2(R2), and ConPhyMP / Journal of Ethnopharmacology best-practice reporting.
