Steps to reproduce

Human induced pluripotent stem cells (hiPSC)-derived trophoblasts into a multi-well microfluidic device to develop a physiologically relevant and scalable placental barrier model. Cultured hiPSC-trophoblasts in 2D or in a perfused 3D micro-channel against a collagen-based matrix. Examined hiPSC-derived trophoblasts behaviour to self-arranged into a 3D structure showing invasive behaviour, fusogenic and endocrine activities, structural integrity and expressing placental transporters. Conducted RNAseq analysis to assesss expression between 2D and 3D, as well as differentation over period of time.
