X. dendrorhous and biosynthetic carotenoid mutants were cultivated in YPD for seven days and imaged with transmission electron microscopy.

Steps to reproduce
Cells were washed first fixed in glutaraldehyde and formaldehyde, then in calcium chloride and post-fixed in osmium tetroxide. The ifxed cells were stained with uranyl acetate solution and embedded in Spurr resin. Sections of resin blocks were stained by toluidine blue and placed over copper/palladium grids. Grids were stained with Reynolds lead under minimal carbon dioxide conditions and viewed on a transmission electron microscope (FEI Tecnai 12) with a digital camera (SIS Morada G2).
