Table of OTUs in Different Saline-Alkali Soils After Exposure to Microplastic Pollution at Various Concentrations
Description
This OTU table was derived from 16S rRNA high-throughput sequencing of alkaline soil samples contaminated with polyethylene microplastics (PE MPs) across varying salinity-alkalinity gradients. The dataset comprises five salinity-alkalinity levels (S1–S5, in increasing order) and three microplastic treatment levels: CK (0% PE MPs), T1 (1% PE MPs), and T2 (5% PE MPs). Preliminary analysis indicates that following the addition of PE MPs, species richness and diversity indices initially decreased and then increased as salinity-alkalinity levels rose. This table is available for other researchers to conduct Alpha/Beta diversity comparisons, Linear Discriminant Analysis Effect Size (LEfSe) for differential species analysis, and functional gene prediction, aiming to further elucidate the ecological risks of combined pollutants in extreme soil environments.

Steps to reproduce
Total genomic DNA was extracted from soil samples using the TGuide S96 Magnetic Soil DNA Kit (Tennen Biologicals, Beijing, China).The bacterial 16S rRNA gene V3-V4 hypervariable region was amplified using the primer pair 338F (5'-ACTCCTACGGGGAGGCAGCA-3') and 806R (5'-GGACTACHVGGGTWTCTAAT-3'). PCR products were examined on agarose gels and purified by Omega DNA purification kit (Omega Inc., Norcross, GA, USA). Purified PCR products were collected and subjected to paired ends (2 × 250 bp) on the Illumina Novaseq 6000 platform. Depending on the quality of single nucleotide, raw data was mainly filtered by Trimmomatic (version 0.33). Primer sequences were identified and removed using Cutadapt (version 1.9.1). The PE reads obtained in the previous steps were assembled with USEARCH (version 10) and then the chimeras were removed with UCHIME (version 8.1). The above steps produce high quality readings for the following analyses. Sequences with >97% similarity were clustered into the same operational taxonomic unit (OTU) by USEARCH (v10), and OTUs with counts less than 2 in all samples were filtered.
