Slides coated with Hep-2 were fixed by 4% PFA, permeabilized by 0.2% Triton, and incubated overnight in a moist chamber at 4℃ with 0.1mL enriched antibodies at a concentration of 100 ug/ml, then were washed in PBS, and incubated for 1h with Alexa Fluor 555-labeled goat antibody to human IgG (Invitrogen). Samples were examined on a confocal microscope (Nikon A1R). Positive staining was determined by comparison with the control samples assessed at equal exposure times and settings.
