This dataset contains the Operational Taxonomic Unit (OTU) table generated from environmental DNA (eDNA) metabarcoding analyses of water samples collected from the Negombo and Puttalam lagoons, Sri Lanka. The table includes OTU identifiers, taxonomic assignments, and read counts for each sample. The dataset provides a resource for assessing aquatic biodiversity, evaluating community composition, and supporting future ecological and conservation studies in Sri Lanka's coastal lagoon ecosystems.

Steps to reproduce

Water samples were collected from selected sites in the Negombo and Puttalam lagoons, Sri Lanka, using an on-site filtration method. Environmental DNA (eDNA) was extracted from the filters, followed by PCR amplification of the target marker gene using metabarcoding primers. The amplified products were sequenced using a high-throughput sequencing platform. Raw sequence data were processed through a bioinformatics pipeline that included quality filtering, primer trimming, chimera removal, sequence clustering into Operational Taxonomic Units (OTUs), and taxonomic assignment using a reference database. The resulting OTU table was generated by summarizing the taxonomic assignments and read counts for each sample and is provided in this dataset.
