This semi-automated nuclei counting method gives the user an opportunity to qualitatively review output results for accuracy. The method was designed to work with fluorescently stained nuclei. If desired, the macro could be modified or applied to other staining methods. Although there are a variety of possible methods to count nuclei (e.g. machine learning), the goal of the method presented here was to run quickly and accurately on a basic consumer-level computer with freely available software (ImageJ).

Data are presented as the Mean ± SEM and statistical significances of differences between two groups were analyzed with the two-side Student’s t-test. One-way ANOVA with the bonferroni post hoc test were used in more than two groups by using GraphPad prism software (La Jolla, CA).
